Afterwards, he took an almost decade long break from acting to pursue studies. Clark met actor Michael Cera while attending college in Los Angeles and the two would eventually become best friends.

They created the series Clark and Michael in which both of them play fictional versions of themselves; the series was directed by Duke and presented during their college thesis at Loyola Marymount University, and the show would eventually be picked up by CBS for a web series.

Clark started to gain popularity through it, and then became a regular in the series Greek as the Christian dorm roommate Dale. He then appeared in Superbad in a small role, and in 2008 became part of the Drunk History Volume 2 alongside Jack Black, with Michael Cera appearing during the first volume.

Duke became the co-star of his first major motion picture, Sex Drive – based on the novel entitled All the Way – which is about a high school graduate who tries to hook up with a girl he met online, and goes on a cross-country road trip with friends.

He was then cast in the film A Thousand Words alongside Eddie Murphy though it remained unreleased until 2012. He was also a part of Kick-Ass which was an independent film co-produced with Brad Pitt and based on the comic book of the same name.

He was then cast in Hot Tub Time Machine playing the role of the nephew of John Cusack’s character. In 2010, he made a cameo in the music video Erase Me by Kid Cudi.
